ET-1 (endothelin-1) regulates a diverse array of physiological processes, including vasoconstriction, angiogenesis and cell proliferation, and most of its effects are associated with an increase in intracellular calcium concentrations. In this Review, Nathan Tykocki and Stephanie Watts describe the interactions between ET-1 and calcium, explore novel mechanisms involved in ET-1-mediated calcium signalling and outline some of the challenges ahead in this area of research.
